Adaptive Multiple Testing Procedure for Clinical Trials with Urn Allocation

چکیده
This work combines the Urn allocation and O’Brien Fleming multiple testing procedure to compare two treatments in clinical trials a novel way. It is shown that this approach overcomes constraints previously made it challenging apply original adaptive design trials. The method provides unique flexibility, enabling be stopped early if one treatment shows superior without compromising efficiency of terms type I error rate power. Experimental data simulated case examples are used illustrate efficacy robustness its potential for usage variety settings.

Adaptive designs for clinical trials with multiple endpoints
Rationale for adaptive designs with multiple endpoints In clinical trials, the efficacy of a new treatment is usually assessed by a single primary endpoint [1,2]. Other important aspects concerning the new treatment, such as adverse events or quality of life, are taken into account by the definition of secondary endpoints. Usually, only the primary endpoint is analyzed confirmatorily with a sta...
متن کاملA Generalized Urn Model in Clinical Trials
Urn models are very popular and useful for adaptive designs in clinical studies. Among various urn models, the drop-the-loser rule is an efficient adaptive treatment allocation scheme, recently proposed for comparing different treatments in clinical trials. This rule is superior to other randomization schemes in terms of variability and power. In this paper, the drop-the-loser rule is generaliz...
متن کاملUsing Urn Models for the Design of Clinical Trials
The essential feature of controlled clinical trials is the random assignment of subjects to two or more treatment groups under investigation. The principle of randomization provides a protection against hidden biases and thus increases the validity of the trial’s findings. Another fundamental aspect of clinical trials is the incorporation into the design and/or analysis those prognostic factors...
متن کامل6 Adaptive Allocation Theory in Clinical Trials – A Review ∗
Various adaptive randomization procedures (adaptive designs) have been proposed to clinical trials. This paper discusses several broad families of procedures, such as the play-the-winner rule and Markov chain model, randomized play-the-winner rule and urn models, drop-the-loser rule, doubly biased coin adaptive design. Asymptotic theories are presented with several pivotal proofs. The effect of...
متن کاملCovariate-Balanced Response-Adaptive Designs for Clinical Trials with Continuous Responses that Target Allocation Proportions
We provide adaptive designs for the sequential allocation of treatments to patients in a clinical trial when the responses are approximately normally distributed. The skewing of the allocations towards the better treatments depends on target proportions for the ranked treatment effects. In addition we combine balance across prognostic factors with a controllable amount of randomization. The asy...
